    Communicating and accentuating the aesthetic and expressive dimension in choral conducting

    This article considers the issues that are involved in effective choral conducting from an aesthetic dimension. Drawing upon research, theories and practice, it provides some insight into the nature of communication and the significance of gesture on vocal outcome as well as qualities of leadership concomitant with such musical activity. The article also reports on a research study that investigated the professional development of students and teachers in the area of choral conducting, focusing on their attitudes, skill acquisition and the importance attached to reflection on practice. The findings reveal that consideration of what counts as effective conducting gesture and communication skill can promote better conducting and, consequently, better, more expressive singing. In addition, the positive impact of self and peer reflection on progress (both face-to-face and within a virtual learning environment) was also acknowledged. Certain suggestions for promoting effective musical leadership in the area of choral conducting are provided, in order to ground theoretical perspectives in practice

    Real time and virtual: tracking the professional development and reflections of choral conductors

    Professional development in conducting is inconsistent across the U.K., leading to a lack of confidence amongst teachers. This paper outlines the continued tracking of the professional development of choral conductors in their skills, knowledge and perceptions of their own progress. The course Choral Conducting, Leadership and Communication takes place over some five months during the spring and summer terms at the Institute of Education, University of London. The students on the course are a mix of i) those on a one year postgraduate initial teacher education programme (PGCE), ii) full- or part-time students on an MA in Music Education Programme and iii) those serving teachers who take it as a non-award bearing CPD course. Students attend four one day practical conducting workshop seminars and are also required to study on-line in a virtual learning environment in the intervening periods. Here they reflect on practice during the face-to-face teaching sessions and in their own professional context through up-loaded video extracts. In addition they retrieve other appropriate study materials and are required to peer assess and form small support groups in discussion forums. This paper describes the progressive development of inclusion of a virtual learning environment (over the period of two years) and the outcomes of this innovative approach to teaching choral conducting which supports face to face teaching

    A more comprehensive and commanding delineation: Mary Shelley's narrative strategy in Frankenstein

    This thesis argues that the first edition of Frankenstein challenges conventional reading by employing what Simpson in Irony and Authority in Romantic Poetry calls Romantic irony, where the absence of a stable 'metacomment' precludes an authoritative reading. The novel hints at such readings but prevents them. The insights offered by Tropp's Mary Shelley's Monster, Baldick's In Frankenstein's Shadow, Poovey's The Proper Lady and the woman writer and Swingle's, 'Frankenstein's Monster and its Relatives: Problems of Knowledge in English Romanticism' are considered, but none recognises the full implications of the instability deriving from multiple first- person narratives. Clemit's The Godwinian Navel acknowledges the novel's indeterminacy, but reads a specific ideological purpose in it. Paradise Last provides a language to describe the relationship between the monster and Frankenstein, but proves too unstable to fix identity or establish moral value. Similarly, Necessity ultimately fails to provide a stable explanation in terms of cause and effect. The status of nature shifts between foreground and background, never allowing final definition. These uncertainties destabilise knowledge which is compromised by its provisional nature: no authoritative reading is possible, yet the novel has narrative coherence. The reader is encouraged to try to develop a reading the structure prevents. The radical nature of the first edition is highlighted by comparison with the 1831 edition, which removes much of the ambivalence and gives the novel a clearer morality. The novel challenges conventional methods of deriving authority by disturbing the reader's orthodox orientation in the world around him' (Simpson) in order to afford 'a point of view to the imagination for the delineation of human passions more comprehensive and commanding than any which the ordinary relations of existing events can yield' (Mary Shelley)

    Phylogenetic determinants of toxin gene distribution in genomes of Brevibacillus laterosporus

    Brevibacillus laterosporus is a globally ubiquitous, spore forming bacterium, strains of which have shown toxic activity against invertebrates and microbes and several have been patented due to their commercial potential. Relatively little is known about this bacterium. Here, we examined the genomes of six published and five newly determined genomes of B. laterosporus, with an emphasis on the relationships between known and putative toxin encoding genes, as well as the phylogenetic relationships between strains. Phylogenetically, strain relationships are similar using average nucleotide identity (ANI) values and multi-gene approaches, although PacBio sequencing revealed multiple copies of the 16S rDNA gene which lessened utility at the strain level. Based on ANI values, the New Zealand isolates were distant from other isolates and may represent a new species. While all of the genomes examined shared some putative toxicity or virulence related proteins, many specific genes were only present in a subset of strains

    Vitamins D2 and D3 Have Overlapping But Different Effects on the Human Immune System Revealed Through Analysis of the Blood Transcriptome

    Vitamin D is best known for its role in maintaining bone health and calcium homeostasis. However, it also exerts a broad range of extra-skeletal effects on cellular physiology and on the immune system. Vitamins D(2) and D(3) share a high degree of structural similarity. Functional equivalence in their vitamin D-dependent effects on human physiology is usually assumed but has in fact not been well defined experimentally. In this study we seek to redress the gap in knowledge by undertaking an in-depth examination of changes in the human blood transcriptome following supplementation with physiological doses of vitamin D(2) and D(3). Our work extends a previously published randomized placebo-controlled trial that recruited healthy white European and South Asian women who were given 15 µg of vitamin D(2) or D(3) daily over 12 weeks in wintertime in the UK (Nov-Mar) by additionally determining changes in the blood transcriptome over the intervention period using microarrays. An integrated comparison of the results defines both the effect of vitamin D(3) or D(2) on gene expression, and any influence of ethnic background. An important aspect of this analysis was the focus on the changes in expression from baseline to the 12-week endpoint of treatment within each individual, harnessing the longitudinal design of the study. Whilst overlap in the repertoire of differentially expressed genes was present in the D(2) or D(3)-dependent effects identified, most changes were specific to either one vitamin or the other. The data also pointed to the possibility of ethnic differences in the responses. Notably, following vitamin D(3) supplementation, the majority of changes in gene expression reflected a down-regulation in the activity of genes, many encoding pathways of the innate and adaptive immune systems, potentially shifting the immune system to a more tolerogenic status. Surprisingly, gene expression associated with type I and type II interferon activity, critical to the innate response to bacterial and viral infections, differed following supplementation with either vitamin D(2) or vitamin D(3), with only vitamin D(3) having a stimulatory effect. This study suggests that further investigation of the respective physiological roles of vitamin D(2) and vitamin D(3) is warranted

    p-tau Ser356 is associated with Alzheimer's disease pathology and is lowered in brain slice cultures using the NUAK inhibitor WZ4003

    Tau hyperphosphorylation and aggregation is a common feature of many dementia-causing neurodegenerative diseases. Tau can be phosphorylated at up to 85 different sites, and there is increasing interest in whether tau phosphorylation at specific epitopes, by specific kinases, plays an important role in disease progression. The AMP-activated protein kinase (AMPK)-related enzyme NUAK1 has been identified as a potential mediator of tau pathology, whereby NUAK1-mediated phosphorylation of tau at Ser356 prevents the degradation of tau by the proteasome, further exacerbating tau hyperphosphorylation and accumulation. This study provides a detailed characterisation of the association of p-tau Ser356 with progression of Alzheimer's disease pathology, identifying a Braak stage-dependent increase in p-tau Ser356 protein levels and an almost ubiquitous presence in neurofibrillary tangles. We also demonstrate, using sub-diffraction-limit resolution array tomography imaging, that p-tau Ser356 co-localises with synapses in AD postmortem brain tissue, increasing evidence that this form of tau may play important roles in AD progression. To assess the potential impacts of pharmacological NUAK inhibition in an ex vivo system that retains multiple cell types and brain-relevant neuronal architecture, we treated postnatal mouse organotypic brain slice cultures from wildtype or APP/PS1 littermates with the commercially available NUAK1/2 inhibitor WZ4003. Whilst there were no genotype-specific effects, we found that WZ4003 results in a culture-phase-dependent loss of total tau and p-tau Ser356, which corresponds with a reduction in neuronal and synaptic proteins. By contrast, application of WZ4003 to live human brain slice cultures results in a specific lowering of p-tau Ser356, alongside increased neuronal tubulin protein. This work identifies differential responses of postnatal mouse organotypic brain slice cultures and adult human brain slice cultures to NUAK1 inhibition that will be important to consider in future work developing tau-targeting therapeutics for human disease.</p

    Fusing actigraphy signals for outpatient monitoring

    [EN] Actigraphy devices have been successfully used as effective tools in the treatment of diseases such as sleep disorders or major depression. Although several efforts have been made in recent years to develop smaller and more portable devices, the features necessary for the continuous monitoring of outpatients require a less intrusive, obstructive and stigmatizing acquisition system. A useful strategy to overcome these limitations is based on adapting the monitoring system to the patient lifestyle and behavior by providing sets of different sensors that can be worn simultaneously or alternatively. This strategy offers to the patient the option of using one device or other according to his/her particular preferences. However this strategy requires a robust multi-sensor fusion methodology capable of taking maximum profit from all of the recorded information. With this aim, this study proposes two actigraphy fusion models including centralized and distributed architectures based on artificial neural networks. These novel fusion methods were tested both on synthetic datasets and real datasets, providing a parametric characterization of the models' behavior, and yielding results based on real case applications. The results obtained using both proposed fusion models exhibit good performance in terms of robustness to signal degradation, as well as a good behavior in terms of the dependence of signal quality on the number of signals fused. The distributed and centralized fusion methods reduce the mean averaged error of the original signals to 44% and 46% respectively when using simulated datasets. The proposed methods may therefore facilitate a less intrusive and more dependable way of acquiring valuable monitoring information from outpatients.This work was partially funded by the European Commission: Help4Mood (Contract No. FP7-ICT-2009-4: 248765).
